| Summary: | The traditional method to propagate the paddy crop are have less percentage to produce a good quality of paddy crop due to factor abiotic and biotic stress that often face breeding problem and complication on incompatibility parental genotype. As a result, the biotechnologies have developed a new transformation to improve the productivity and quality of the paddy yield through in vitro propagation method. The aim for this experiment for this research project is to determine the different concentration of 2, 4- Dichlorophenoxyacetic acid (2, 4- D) towards callus induction and growth performance of paddy (Oryza sativa L.) where it is important in biotechnology and plant regeneration purposed. It is important to measure the different concentration of (2,4- D) hormone towards weight of callus induction and to identify the effect of different concentration of (2, 4- D) towards growth performance of paddy plant (Oryza sativa L.).From this experiment it can help the producer and researcher to regenerate and culture the paddy plant with less seed used. Other than that, it also were reduced the potential of infection from pest and disease that normally happened on the field due to control environmental condition. Mature paddy seed was culture in MS media that supplemented with different concentration of (2, 4-D) (1.0 mg/L, 0.5 mg/L, 1.5 mg/L, 0.0 mg/L and no treatment) combination with constant NAA and BAP for every treatment. A Complete Randomized Design (CRD) with 5 replications was used in this study. The result showed, the higher concentration of (2, 4-D) (1.5 mg/L) with combination of NAA and BAP are the highest of weight of callus produced .The increased of (2, 4-D) on the medium were lead to the increase of the weight of callus but it not suitable used to support the growth performance of paddy plant arise from mature paddy seed. The growth performance of plantlet is improved by week but the improvement is not significant until the end of studied. |
